I always recommend for owners that they should not use any medicine in their pets without consulting a vet because medicines either over the counter or prescription drugs can be toxic for dogs. Benadryl is used to treat allergy and FDA has not approved this drug for animal use. Vets can prescribe this medicines in animals because they know about side effects and dose.

Oral dose of the Benadryl in dogs is 0.5-2 mg per pound body weight for 2-3 times in a day. For your Yorkie of 5 pounds, its dose can be 2.5-10 mg for 2-3 times a day.

WikiAnswers says that a vet recommended up to 1mg. Per pound every 8 hours. So since your dog is 5 lbs, that would be 5mg. Every 8 hours. It also says that it's not recommended if your dog has prostate disease, hyperthyroidism or glaucoma. Of course it also says that when giving any med to humans or animals, it's strongly advised that you get professional advice from your doctor or a vet.
