I would say elephants are the biggest (no pun intended) enemies of the lion. Also I'm sure man is in there somewhere as well...

Leopards, Cheetahs, and Hyaenas
Man, other lions, hyaenas. Water buffalo have also been known to kill lions in defence of themselves and their families. (But they don't pick a fight upfront :) )
